Human MutL Homolog 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com designed for the in vitro quantitative determination of human MLH1 in tissue homogenates, cell lysates or other biological fluids. The assay uses a pre-coated 96-well microplate to which an antibody specific for MLH1 has been immobilised. Standards and test samples are added to the wells, and MLH1 present in each sample is bound by the immobilised antibody. After washing, a Biotinylated Anti-MLH1 Antibody binds the captured MLH1, followed by addition of an HRP-Avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue colour that changes to yellow upon addition of the acidic stop solution. The density of yellow is proportional to the amount of MLH1 captured, and the concentration is calculated from the O.D. absorbance read at 450 nm in a microplate reader.

The assay has a reported sensitivity of 0.057 ng/ml and a working range of 0.156–10 ng/ml. Reported recovery values (n=5) average 91% in serum (range 80–102%), 90% in EDTA plasma (range 81–100%), and 84% in heparin plasma (range 80–89%). The total assay time is approximately 4 hours 30 minutes. Kit components should be stored at +4°C or -20°C as instructed, and the product is intended for research use only.
