Human Muscle, Skeletal, Receptor Tyrosine Kinase ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Muscle, Skeletal, Receptor Tyrosine Kinase (MUSK) in tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a pre-coated microplate with colorimetric detection, and absorbance is read at 450 nm on a microplate reader. Reactivity is confirmed for human samples.

The quantitative range is 0.156–10 ng/ml, with a reported sensitivity of 0.054 ng/ml and a total assay time of 4 hours 30 minutes. Validation studies reported the following recovery values: serum (n=5) ranged from 80% to 102% with an average of 91%; EDTA plasma (n=5) ranged from 81% to 100% with an average of 90%; and heparin plasma (n=5) ranged from 80% to 89% with an average of 84%. Dilution linearity assessed at 1:2, 1:4, 1:8 and 1:16 yielded recoveries of 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.

In the workflow, standards and samples are added to the wells and MUSK is bound by the immobilised capture antibody. After washing, a biotinylated anti-MUSK detection antibody is added, followed by HRP-Avidin conjugate. TMB substrate is then added to produce a blue colour that turns yellow upon addition of acidic stop solution; the intensity of the yellow colour is proportional to the amount of MUSK captured in each well.
