Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit from Antibodies.com for the in vitro quantitative determination of human Mucosal Addressin Cell Adhesion Molecule 1 (MAdCAM1). The assay is intended for use with serum, plasma, tissue homogenates, cell lysates, urine and other biological fluids, and is reactive with human samples.

The kit uses a 96-well microplate pre-coated with an antibody specific for MAdCAM1. Standards and samples are added to the wells, where MAdCAM1 is captured by the immobilised antibody. After washing, a biotinylated anti-MAdCAM1 detection antibody is added, followed by an HRP-avidin conjugate and TMB substrate. The enzymatic reaction produces a blue product that turns yellow upon addition of an acidic stop solution; absorbance is read at 450 nm using a colorimetric microplate reader, and the concentration of MAdCAM1 is calculated by reference to a standard curve. The total assay time is 4 hours 30 minutes.

The product range is 0.312–20 ng/ml with a sensitivity of 0.125 ng/ml. In spike-and-recovery testing (n=5), average recoveries were 91% in serum (range 80–102%), 91% in EDTA plasma (range 81–101%) and 84% in heparin plasma (range 80–89%). Linearity upon dilution across 1:2 to 1:16 (n=5) was 74–107% in serum, 79–108% in EDTA plasma and 82–105% in heparin plasma. Kit components should be stored at +4 °C or −20 °C as instructed, and the kit should not be used past its expiration date.
