Human Mucin 6 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com, designed for the in vitro quantitative determination of human Gastric Mucin/MUC-6 in serum, plasma, tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, in which an antibody specific for Gastric Mucin/MUC-6 has been pre-coated onto a 96-well microtiter plate. Standards and test samples are added and Gastric Mucin/MUC-6 present in each sample is captured by the immobilised antibody. After washing, a Biotinylated Anti-Gastric Mucin/MUC-6 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then added; HRP catalysis produces a blue-coloured product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of target captured, and concentration is calculated by reading O.D. absorbance at 450 nm in a microplate reader and referring to a standard curve.

Reported specifications include reactivity against human Gastric Mucin/MUC-6, a sensitivity of 0.115 ng/ml, an assay range of 0.312–20 ng/ml and an assay time of 4h 30m. Detection is colourimetric, the platform is a pre-coated microplate of 12 x 8 well strips, and components are stored at +4°C or −20°C as instructed; the kit should not be used past the stated expiration date.

Spike-and-recovery performance (n=5) is reported as an average of 91% (range 80–102%) for serum, 90% (range 81–100%) for EDTA plasma and 84% (range 80–89%) for heparin plasma. Dilution linearity is also provided at 1:2, 1:4, 1:8 and 1:16 dilutions across serum, EDTA plasma and heparin plasma.
