Human Mucin 4 ELISA Kit

A sandwich ELISA from Antibodies.com designed for the in vitro quantitative determination of human Mucin 4 (MUC4) in serum, plasma, tissue homogenates or other biological fluids. The assay employs a pre-coated microplate (12 x 8 well strips) onto which an antibody specific for MUC4 has been immobilised. Standards and test samples are added to the wells, and any MUC4 present is captured by the coated antibody. After washing, a Biotinylated Anti-MUC4 Antibody is added, followed by an HRP-Avidin conjugate. Following incubation and further washing, TMB substrate solution is added; catalysis produces a blue product that turns yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of MUC4 captured, and the concentration is calculated by reading absorbance at 450 nm on a colorimetric microplate reader against the standard curve.

The kit has a stated sensitivity of 0.115 ng/ml and an assay range of 0.312–20 ng/ml, with a total assay time of approximately 4 h 30 min. Detection is colorimetric, and the kit is reactive with human samples. Kit components should be stored at +4 °C or −20 °C as instructed, and the kit must not be used past its expiration date.

Reported recovery data fall within 80–102% for serum (average 91%), 81–100% for EDTA plasma (average 90%), and 80–89% for heparin plasma (average 84%). Dilution linearity at 1:2 to 1:16 is also provided for each sample matrix, supporting use with serum, plasma and tissue homogenates.
