Description

From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Mucin 17 (MUC17) in serum, plasma, tissue homogenates, and other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for MUC17, and the target is quantified by reading the optical density at 450 nm against a standard curve. Reported sensitivity is 6.0 pg/ml, with a working standard range of 15.6-1000 pg/ml. Detection is colourimetric, employing an HRP-Avidin conjugate and TMB substrate; the enzymatic reaction produces a blue product that changes to yellow upon addition of acidic stop solution, with intensity proportional to MUC17 concentration.

The procedure follows a standard sandwich ELISA workflow: standards and samples are added to pre-coated wells and incubated, followed by sequential incubation with a biotinylated anti-MUC17 detection antibody and HRP-Avidin conjugate, with washes between steps. The kit has been validated on serum, EDTA plasma and heparin plasma samples, with mean recoveries of 91%, 96% and 95% respectively. Serum samples are collected in serum separator tubes, plasma with EDTA or heparin as anticoagulant, and tissues are homogenised in PBS prior to assay; samples should be centrifuged, aliquoted and stored to avoid repeated freeze/thaw cycles. Reagents are stored at +4°C or -20°C as instructed and the product is intended for research use only, not for diagnostic or therapeutic applications.
