The assay employs a pre-coated 96-well microplate (12 x 8 well strips) bearing a Smad4-specific capture antibody, followed by a biotinylated anti-Smad4 detection antibody, an HRP-avidin conjugate, and TMB substrate; the enzymatic reaction is stopped with an acidic solution and absorbance is read at 450 nm on a colorimetric microplate reader, with concentration determined against a standard curve.

According to the manufacturer's specification, sensitivity is <0.112 ng/ml, the assay range is 0.312–20 ng/ml, and total assay time is 4h 30m. Sample recovery testing (n=5) gave averages of 91% for serum (range 84–99%), 92% for EDTA plasma (range 82–103%), and 93% for heparin plasma (range 84–102%). Dilution linearity observed across 1:2 to 1:16 sample dilutions was 72–106% for serum, 84–110% for EDTA plasma, and 74–106% for heparin plasma.
