Description
Manufactured by antibodies.com, this Human Smad3 (Mothers Against Decapentaplegic Homolog 3) ELISA kit is a sandwich enzyme-linked immunosorbent assay (sELISA) intended for the in vitro quantitative determination of human Smad3 in tissue homogenates, cell lysates, or other biological fluids. The assay is reactive with human samples and uses a pre-coated microplate platform with colourimetric detection read on a microplate reader at 450 nm. The reported assay range is 0.156–10 ng/ml with a sensitivity of <0.066 ng/ml, and the total assay time is 4 h 30 min.
The procedure uses an antibody specific for Smad3 pre-coated onto the plate wells; standards and samples are added and Smad3 is captured, followed by incubation with a biotinylated anti-Smad3 antibody, HRP-avidin conjugate, and TMB substrate. The reaction produces a blue product that turns yellow upon addition of the acidic stop solution, with absorbance proportional to Smad3 concentration. Reported recovery values were: serum (n=5) 84–99% (average 91%), EDTA plasma (n=5) 82–103% (average 92%), and heparin plasma (n=5) 84–102% (average 93%).
Kit components include a pre-coated 96 well microplate, lyophilized standard, Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers. Components are stored at +4°C or −20°C as indicated, and the kit should not be used past its expiration date.
