Human Mothers Against Decapentaplegic Homolog 1 ELISA Kit

From Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of human Smad1 in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The kit is intended for use with a colorimetric microplate reader and is reactive with human samples.

Assay performance is defined by a product range of 0.781–50 ng/ml and a sensitivity of <0.33 ng/ml, with results generated through a 4h 30m protocol. The procedure uses a pre-coated 96-well microplate (12 x 8 well strips) on which standards and samples are captured by an immobilized anti-Smad1 antibody, then detected using a Biotinylated Anti-Smad1 Antibody, HRP-Avidin conjugate and TMB substrate. The resulting absorbance is read at 450 nm against a standard curve. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.

Validation data report recovery averages of 91% for serum (range 84%–99%, n=5), 92% for EDTA plasma (range 82%–103%, n=5) and 93% for heparin plasma (range 84%–102%, n=5). Dilution linearity across 1:2 to 1:16 dilutions gave recoveries of 72%–106% for serum, 84%–110% for EDTA plasma and 74%–106% for heparin plasma.
