Description

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human CXCL9 (Monokine Induced By Interferon Gamma) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids.

The assay employs a sandwich enzyme immunoassay technique on a pre-coated 96-well microplate configured as 12 x 8 well strips. Standards and test samples are added to the wells, where CXCL9 is captured by the immobilised antibody. After incubation and washing, a Biotinylated Anti-CXCL9 Antibody and HRP-Avidin conjugate are added sequentially. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of CXCL9 captured, and concentration is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader and referring to a standard curve. The kit has a reported product range of 0.156-10 ng/ml and a sensitivity of <0.062 ng/ml, with a total assay time of 4h 30m. Kit components include a pre-coated 96 well microplate, lyophilised standard, Detection Solution A, Detection Solution B, Wash Buffer 30X, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, stored at +4°C or -20°C as instructed.

Reported sample recovery values (average, n=5) are 91% in serum (range 80-102%), 90% in EDTA plasma (81-100%) and 84% in heparin plasma (80-89%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 dilutions is given as 87-91%, 87-107%, 74-101% and 92-97% respectively for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma.
