Description

Available from Antibodies.com as part of the Assays and Kits range, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is designed for the in vitro quantitative determination of human Metanephrine (MN). It is suitable for use with serum, plasma, tissue homogenates, and other biological fluids, and has been validated for reactivity with human samples.

The 2-hour (120 minute) assay is performed on a pre-coated 96-well microplate and read colorimetrically at 450 nm using a standard microplate reader. Metanephrine present in standards or samples competes with the immobilized coating for binding to an assay-specific antibody. After washing, an HRP-Streptavidin conjugate is added, followed by TMB substrate solution, which produces a blue-coloured product that turns yellow on addition of acidic stop solution. The OD450 reading is inversely proportional to metanephrine concentration in the sample and is interpolated from a standard curve.

Quoted performance figures include a sensitivity of <9.375 pg/mL and a working range of 15.625–1000 pg/mL. The antibody is specific for Metanephrine, with no reported cross-reactivity with other analogs or variants. Recovery has been characterised in serum (88–103%, average 97%, n=10), EDTA plasma (88–103%, average 96%, n=10) and heparin plasma (86–104%, average 93%, n=10), and dilution linearity has been demonstrated at 1:2, 1:4 and 1:8 dilutions. All reagents are shipped at +4°C and stored at +4°C or −20°C as directed in the protocol manual; the kit must not be used past its expiration date and freeze/thaw cycles should be avoided.
