Human MMP13 ELISA Kit

The kit is suitable for serum and plasma samples, and also for tissue homogenates and other biological fluids. It is reported to be specific for MMP13 with no reported cross-reactivity with other analogs or variants. The manufacturer reports a sensitivity of less than 5 pg/ml and an assay range of 78-5000 pg/ml, and describes the protocol as easy to follow.

Running in approximately 4 hours, the assay uses a pre-coated microplate platform with colorimetric detection read at 450 nm. Standards and samples bind to the immobilized capture antibody; a biotinylated detection antibody and HRP-Streptavidin conjugate are then added, and TMB substrate produces a blue colour that turns yellow upon addition of acidic stop solution. The intensity of the yellow colour is directly proportional to the amount of MMP13 captured, and the concentration is calculated from a standard curve. The kit is shipped at +4°C, with components stored at +4°C or -20°C as instructed; freeze/thaw cycles should be avoided and the kit should not be used past its expiration date.

Serum and plasma samples may affect test results and should be diluted at least 1:2 with Sample Dilution Buffer prior to testing. Validated sample dilutions include healthy serum at 1:2 and U2OS cell culture supernatants used undiluted. Reported recovery values from sample testing include serum (85% - 103%, average 95%, n=10), EDTA plasma (91% - 104%, average 97%, n=10) and heparin plasma (97% - 104%, average 99%, n=10).
