Human Minichromosome Maintenance Deficient 5 ELISA Kit

From Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of human MCM5 (Minichromosome Maintenance Deficient 5) in tissue homogenates or other biological fluids. It uses a pre-coated microplate format with colorimetric detection read at 450 nm in a microplate reader. The assay is specific for human samples and reports a sensitivity of <0.053 ng/ml with a working range of 0.156–10 ng/ml. The full protocol takes 4h 30m.

The workflow uses a biotinylated anti-MCM5 detection antibody and an HRP-avidin conjugate. After incubation with TMB substrate, addition of the acidic stop solution yields a yellow product whose absorbance is proportional to the MCM5 concentration in each well. Supplied components include a pre-coated microplate, lyophilised standard, Detection Solutions A and B, wash and sample dilution buffers, TMB substrate, stop solution, and adhesive plate sealers, with storage at +4°C or –20°C depending on the component.

Spike-and-recovery performance has been validated for serum, EDTA plasma, and heparin plasma (n=5 per matrix). Average recoveries were 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%), and 84% for heparin plasma (range 80–89%). Dilution linearity has been demonstrated at 1:2, 1:4, 1:8, and 1:16 across the same three matrices.
