From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit is designed for the in vitro quantitative determination of human MCM2 in serum, plasma, tissue homogenates or other biological fluids. The assay employs a pre-coated microplate (12 x 8 well strips) with an antibody specific for MCM2, followed by detection with a Biotinylated Anti-MCM2 Antibody and an HRP-Avidin conjugate. The enzymatic reaction is visualised using TMB substrate, producing a blue product that changes to yellow upon addition of stop solution; absorbance is read at 450 nm in a colorimetric microplate reader and the MCM2 concentration is calculated against a standard curve. Total assay time is 4 hours 30 minutes.

The kit has a sensitivity of <0.058 ng/ml and an assay range of 0.156-10 ng/ml. Reported sample recovery values are 80-102% (average 91%) for serum (n=5), 81-100% (average 90%) for EDTA plasma (n=5), and 80-89% (average 84%) for heparin plasma (n=5). Dilution linearity has been demonstrated across 1:2 to 1:16 in serum (74-107%), EDTA plasma (79-108%) and heparin plasma (82-105%). Kit components are stored at +4°C or -20°C as indicated, and the kit should not be used past its expiration date.
