The microtiter plate is pre-coated with an antibody specific for MFGE8; standards and samples are added, followed by a biotinylated anti-MFGE8 detection antibody, an HRP-avidin conjugate, and a TMB substrate, producing a yellow colour read at 450 nm whose absorbance is proportional to MFGE8 concentration.
