From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) is configured for the in vitro quantitative determination of human Microtubule Associated Protein Tau (Tau) in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay is reactive with human samples and uses a pre-coated 96-well microplate arranged as 12 x 8 well strips.

The procedure employs an immobilized capture antibody specific for Tau, followed by incubation with a Biotinylated Anti-Tau Antibody and then HRP-Avidin conjugate. TMB substrate is added to visualize the reaction; the resulting blue product turns yellow upon addition of acidic stop solution, and absorbance is read at 450 nm on a colorimetric microplate reader. Quantification is achieved by interpolation against a standard curve generated from the lyophilized standards supplied with the kit.

Key performance characteristics include a stated sensitivity of 29 pg/ml and a standard curve range of 78.1-5000 pg/ml, with a total assay time of 4h 30m. Spike-and-recovery data in serum (n=5) covers a range of 80%-102% with an average of 91%; EDTA plasma (n=5) covers 81%-100% (90% average) and heparin plasma (n=5) covers 80%-89% (84% average). Linearity across dilutions from 1:2 to 1:16 is also reported for each matrix. Kit components include the pre-coated microplate, lyophilized standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers, with storage at +4°C or -20°C as indicated per component.
