Description

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com is designed for the in vitro quantitative determination of human Microtubule Associated Protein 4 (MAP4) in tissue homogenates or other biological fluids. The assay uses a pre-coated microplate with an antibody specific for MAP4. Standards and test samples are added to the wells, where MAP4 binds to the immobilised antibody. After incubation and washing, a Biotinylated Anti-MAP4 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of MAP4 captured in each well, and concentration is calculated by reading O.D. absorbance at 450 nm in a microplate reader against a standard curve.

The assay has a reported range of 0.312–20 ng/ml and a sensitivity of <0.101 ng/ml, with a total assay time of 4h 30m. Detection is colourimetric. Recovery testing (n=5) gave averages of 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%), and 84% for heparin plasma (range 80–89%). Linearity was demonstrated across 1:2 to 1:16 dilutions for all three sample types. Kit components should be stored at +4°C or −20°C as instructed.
