From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Microphthalmia Associated Transcription Factor (MiTF) in tissue homogenates, cell lysates, or other biological fluids. The assay uses an antibody specific for MiTF pre-coated onto a 96-well microplate. Standards and samples are added to the wells, followed by a Biotinylated Anti-MiTF Antibody and an HRP-Avidin conjugate. A TMB substrate is then used to visualize the enzymatic reaction; the colour change is read at 450 nm in a colorimetric microplate reader and the concentration of MiTF is calculated by reference to a standard curve.
Reported sensitivity is <0.059 ng/ml with an assay range of 0.156–10 ng/ml and a total assay time of 4 h 30 min. Recovery values across sample matrices are 80–102% (average 91%) for serum, 81–100% (average 90%) for EDTA plasma, and 80–89% (average 84%) for heparin plasma. Kit components should be stored at +4 °C or −20 °C as instructed, and the kit must not be used past its expiration date.
