The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with absorbance read at 450 nm on a colorimetric microplate reader.

The reported assay range is 0.156-10 ng/ml, with a sensitivity of 0.069 ng/ml. Standards and samples are captured by an immobilized antibody specific for MSRA, followed by incubation with a Biotinylated Anti-MSRA Antibody and an HRP-Avidin conjugate. Signal is visualised with TMB substrate, producing a blue product that converts to yellow after addition of acidic stop solution; the density of yellow is proportional to MSRA concentration and is quantified against a standard curve.
