A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com designed for the in vitro quantitative determination of human Metallothionein 1E (MT1E) in serum, plasma, tissue homogenates or other biological fluids. The assay uses an antibody specific for MT1E pre-coated onto a 96-well microplate. Standards and test samples are added to the wells and MT1E present in each sample is bound by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-MT1E Antibody is added, followed by an HRP-Avidin conjugate. After further incubation and washing, TMB substrate solution is used to visualise the enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The intensity of the yellow colour is proportional to the amount of MT1E captured, and the concentration is calculated by reading the absorbance at 450 nm in a microplate reader and referring to the standard curve.
The assay has a sensitivity of <0.119 ng/ml and an assay range of 0.312–20 ng/ml, with a total assay time of 4 hours 30 minutes. Detection is colourimetric on a pre-coated microplate supplied as 12 x 8 well strips. Sample recovery has been characterised: serum (n=5) 83–100% with an average of 91%, EDTA plasma (n=5) 90–103% averaging 96%, and heparin plasma (n=5) 89–102% averaging 95%. Dilution linearity at 1:2, 1:4, 1:8 and 1:16 dilutions has also been evaluated across these sample types.
Kit components include the pre-coated 96 well microplate, lyophilized standard, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Components should be stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date.
