The Antibodies.com Human Membrane Cofactor Protein ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of human CD46 in serum, plasma, urine, saliva and other biological fluids. The assay uses an antibody specific for CD46 pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, where CD46 present in each sample is bound by the immobilised antibody. After incubation and washing, a Biotinylated Anti-CD46 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow colour is proportional to the amount of CD46 captured in each well, and the concentration of CD46 is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The assay has a stated product range of 6.25-400 pg/ml and a sensitivity of 2.34 pg/ml, and is read using a colorimetric microplate reader. Total assay time is 4h 30m. Kit components should be stored at +4°C or -20°C as instructed and must not be used past the stated expiration date. The platform is a pre-coated microplate configured as 12 x 8 well strips, and the kit is reported as suitable for serum, plasma and urine.

Validated sample types include serum, EDTA plasma and heparin plasma. Reported spike-and-recovery data are: average 91% for serum (range 80-102%, n=5), average 90% for EDTA plasma (range 81-100%, n=5) and average 84% for heparin plasma (range 80-89%, n=5). Dilution linearity, reported at dilutions of 1:2, 1:4, 1:8 and 1:16, was 87-91%, 87-107%, 74-101% and 92-97% respectively for serum (n=5); 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma (n=5); and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma (n=5).
