Human Melatonin Receptor 1B ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of human Melatonin Receptor 1B/MTNR1B in tissue homogenates, cell lysates, or other biological fluids. The assay employs the sandwich enzyme immunoassay technique. An antibody specific for Melatonin Receptor 1B/MTNR1B is pre-coated onto a 96-well microplate; standards and test samples are added, and the target in each sample is bound by the immobilized antibody. After washing, wells are incubated with Biotinylated Anti-Melatonin Receptor 1B/MTNR1B Antibody, followed by HRP-Avidin conjugate. Following further incubation and washing, TMB substrate is added; HRP catalysis produces a blue product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Melatonin Receptor 1B/MTNR1B captured, and concentration is calculated from the O.D. absorbance at 450 nm using a standard curve.

The product range is 0.156–10 ng/ml, with a sensitivity of <0.058 ng/ml. The assay is performed on a pre-coated microplate (12 x 8 well strips) and takes 4h 30m to complete. Aggregate range and average values are reported for serum (n=5) 83–100%, average 91%; EDTA plasma (n=5) 90–98%, average 94%; and heparin plasma (n=5) 89–102%, average 95%. Per-sample values are also provided across dilutions of 1:2, 1:4, 1:8 and 1:16 for serum, EDTA plasma, and heparin plasma. The kit is suitable for cell lysates and tissue homogenates. Kit components are stored at +4°C or −20°C as instructed, and the kit must not be used past the stated expiration date.
