Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com enables the in vitro quantitative determination of human Melanoma Inhibitory Activity Protein 1 in serum, plasma or other biological fluids.

An antibody specific for Melanoma Inhibitory Activity Protein 1 is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and test samples are added to the wells, and the target protein present in each sample is bound by the immobilized antibody. After washing, a Biotinylated Anti-Melanoma Inhibitory Activity Protein 1 Antibody is added, followed by HRP-Avidin conjugate. A TMB substrate is then added to visualise the enzymatic reaction, producing a blue-coloured product that turns yellow after addition of acidic stop solution. The intensity of yellow is proportional to the amount of target captured, and concentration is calculated from the absorbance at 450nm against a standard curve.

The assay range is 78.1-5000 pg/ml with a sensitivity of 25 pg/ml. Total assay time is 4 hours 30 minutes, and detection is colorimetric. Recovery data show averages of 91% (serum, n=5, range 80-102%), 90% (EDTA plasma, n=5, range 81-100%) and 84% (heparin plasma, n=5, range 80-89%). Dilutional linearity data across serial dilutions are reported for serum, EDTA plasma and heparin plasma. Kit components include a pre-coated 96-well microplate, lyophilized standard, Detection Solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers, stored at +4°C or -20°C as instructed.
