From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Matrix Metalloproteinase 9 (MMP9). The kit is suitable for serum, plasma, tissue homogenates, cell lysates, cell culture supernates, and other biological fluids, and runs on a pre-coated 96-well microtiter plate arranged as 12 x 8 well strips, read at 450 nm on a colorimetric microplate reader. In the workflow, MMP9 present in standards or samples is captured by an immobilized antibody, followed by binding of a Biotinylated Anti-MMP9 Antibody and an HRP-Avidin conjugate, with TMB substrate producing a blue-to-yellow colour change proportional to MMP9 concentration.

Performance characteristics include a product measuring range of 0.156–10 ng/ml and a sensitivity of <0.063 ng/ml, with a total assay time of 4 hours 30 minutes. Recovery data (n=5) shows 80–102% (average 91%) for serum, 81–100% (average 90%) for EDTA plasma, and 80–89% (average 84%) for Heparin plasma. Linearity values are reported for serum, EDTA plasma, and Heparin plasma at dilutions of 1:2, 1:4, 1:8, and 1:16.

Kit components include a pre-coated 96-well microplate, lyophilized standard, Detection Solution A and Detection Solution B (stored at -20°C), 30X Wash Buffer (stored at +4°C), Sample Dilution Buffer (stored at -20°C), TMB Substrate (stored at +4°C), Stop Solution (stored at +4°C), and plate sealers. Kit components are stored at +4°C or -20°C as instructed, and the product should not be used past its expiration date.
