Human Matrix Metalloproteinase 3 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human MMP3. The kit is suitable for serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids, and uses a pre-coated 96-well microplate (12 x 8 well strips) read at 450 nm on a colorimetric microplate reader.

An antibody specific for MMP3 is pre-coated on the plate; standards and samples are added, followed by a Biotinylated Anti-MMP3 Antibody and HRP-Avidin conjugate. TMB substrate produces a blue product that turns yellow upon addition of stop solution, with absorbance proportional to MMP3 concentration and quantified against a standard curve. Total assay time is 4 hours 30 minutes.

The standard curve spans 0.156–10 ng/ml with a sensitivity of <0.068 ng/ml. Reported recovery in serum (n=5) ranges from 80–102% (average 91%), in EDTA plasma (n=5) from 81–100% (average 90%), and in heparin plasma (n=5) from 80–89% (average 84%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions is 87–91%, 87–107%, 74–101% and 92–97% for serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5). Kit components should be stored at +4°C or −20°C as instructed and not used past the expiration date.
