Human Matrix Metalloproteinase 13 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of human Matrix Metalloproteinase 13 (MMP13) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.

The kit uses a 96-well microtiter plate pre-coated with an antibody specific for MMP13. Standards and samples are added to the wells and the MMP13 present is bound by the immobilized capture antibody. After incubation and washing, a Biotinylated Anti-MMP13 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution visualises the HRP enzymatic reaction by catalysis, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader, and the concentration of MMP13 is calculated by reference to the standard curve.

The product range is 0.156-10 ng/ml and the sensitivity is <0.062 ng/ml, with a total assay time of 4 hours 30 minutes. Recovery in serum (n=5) ranged from 83% to 100% with an average of 91%, in EDTA plasma (n=5) from 90% to 103% averaging 96%, and in heparin plasma (n=5) from 89% to 102% averaging 95%. Linearity was evaluated at dilutions of 1:2, 1:4, 1:8 and 1:16 across serum, EDTA plasma and heparin plasma, each with n=5. Kit components include a pre-coated 96-well microplate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, with storage at +4°C or -20°C as instructed.
