Human Matrix Metalloproteinase 12 ELISA Kit

A sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of human Matrix Metalloproteinase 12 (MMP12) in serum, plasma, or other biological fluids. The assay employs a pre-coated 96-well microplate (12 x 8 well strips), with standards and samples incubated alongside a biotinylated anti-MMP12 detection antibody. An HRP-avidin conjugate and TMB substrate complete the sandwich, yielding a blue product that turns yellow upon addition of an acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and MMP12 concentration is calculated against a standard curve.

The working range is 0.312-20 ng/ml, with a sensitivity of 0.114 ng/ml and a total assay time of approximately 4h 30m. Validation data (n=5) show average recoveries of 91% in serum (range 83-100%), 96% in EDTA plasma (90-103%), and 95% in heparin plasma (89-102%). Linearity was maintained across 1:2, 1:4, 1:8, and 1:16 sample dilutions for serum (79-110%), EDTA plasma (73-109%), and heparin plasma (77-110%). Kit components are stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
