Human Matrix Gla Protein ELISA Kit

From Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of Matrix Gla Protein in human serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips with a colorimetric readout measured at 450 nm in a microplate reader. Standards and samples are added to antibody-coated wells; captured analyte is detected with a biotinylated anti-Mgp antibody followed by an HRP–avidin conjugate and TMB substrate, producing a yellow colour whose intensity is proportional to Matrix Gla Protein concentration. Total assay time is 4 hours 30 minutes.

Performance characteristics include a standard curve range of 15.6–1000 pg/ml and a sensitivity of 6.7 pg/ml. Reported recovery in serum (n=5) is 80%–102% with an average of 91%, in EDTA plasma (n=5) 81%–100% averaging 90%, and in heparin plasma (n=5) 80%–89% averaging 84%. Linearity has been characterised across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma. Supplied components include the pre-coated microplate strips, lyophilized standard, detection solutions, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers.
