Human Mannose Binding Lectin ELISA Kit

From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Mannose Binding Lectin (MBL) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated 96-well microplate format with colorimetric detection. An antibody specific for MBL is immobilised on the plate; standards and samples are added and bound, followed by a biotinylated anti-MBL detection antibody and HRP-Avidin conjugate. TMB substrate is used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of stop solution; the density of yellow, read at 450 nm in a microplate reader, is proportional to the MBL concentration in each well.

Manufacturer-stated performance characteristics include a sensitivity of 0.068 ng/ml and a standard range of 0.156–10 ng/ml. Reported spike-and-recovery averages were 91% in serum (range 80–102%, n=5), 90% in EDTA plasma (range 81–100%, n=5) and 84% in heparin plasma (range 80–89%, n=5). Dilution linearity across 1:2 to 1:16 dilutions was reported at 87–91% to 92–97% for serum, 90–105% to 79–108% for EDTA plasma and 84–95% to 89–91% for heparin plasma. Total assay time is 4h 30m. Kit components are stored at +4°C or –20°C as instructed, and the kit should not be used past its expiration date.
