Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com enables the in vitro quantitative determination of human HLA B in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The kit is reactive with human samples.

The 96-well microplate is pre-coated with an antibody specific for HLA B. Standards and test samples are added to the wells, where HLA B present in each sample is bound by the immobilized antibody. After incubation and washing, a Biotinylated Anti-HLA B Antibody binds the captured HLA B, followed by addition of an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of HLA B captured, measured by absorbance at 450 nm using a microplate reader and compared to a standard curve.

Reported performance includes a product range of 1.56-100 ng/ml and a sensitivity of 0.63 ng/ml. The pre-coated microplate platform is arranged as 12 x 8 well strips and the total assay time is 4h 30m. Detection is colourmetric. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date. Dilution linearity has been demonstrated for serum (87-107% across 1:2 to 1:16 dilutions), EDTA plasma (79-108%) and heparin plasma (82-105%).
