A sandwich enzyme-linked immunosorbent assay (ELISA) from Antibodies.com is designed for the in vitro quantitative determination of human HLA A (Major Histocompatibility Complex Class I A) in tissue homogenates or other biological fluids. The assay uses a pre-coated 96-well microtiter plate to which an antibody specific for HLA A has been immobilised. Standards and test samples are added to the wells, where HLA A is bound by the immobilised antibody. After washing, a biotinylated anti-HLA A antibody is added, followed by an HRP-avidin conjugate. A TMB substrate solution is then used to visualise the reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The concentration of HLA A is calculated by reading the optical density at 450 nm with a colorimetric microplate reader and referring to the standard curve.

The assay has a reported sensitivity of 0.126 ng/ml and a product range of 0.312–20 ng/ml, with a total assay time of 4 hours 30 minutes. Recovery data for serum (n=5) is 80%–102% with an average of 91%; for EDTA plasma (n=5), 81%–100% with an average of 90%; and for heparin plasma (n=5), 80%–89% with an average of 84%. Dilution linearity data are also reported for serum, EDTA plasma, and heparin plasma samples at 1:2, 1:4, 1:8, and 1:16 dilutions. Kit components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.

The kit is intended for use with human samples and is listed under the Assays and Kits category.
