Human Major Basic Protein ELISA Kit

From Antibodies.com, the Human Major Basic Protein ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay designed for the in vitro quantitative determination of human PRG2 (Major Basic Protein) in serum, plasma, tissue homogenates, or other biological fluids. The assay is reactive with human samples and uses a pre-coated 96-well microtiter plate arranged as 12 x 8 well strips.

The kit reports a sensitivity of <0.165 ng/ml and a product range of 0.468-30 ng/ml, with a typical assay time of 4h 30m. In undiluted samples, recovery averaged 91% in serum (range 80%-102%, n=5), 90% in EDTA plasma (81%-100%, n=5), and 84% in heparin plasma (80%-89%, n=5). Dilution linearity was evaluated at 1:2, 1:4, 1:8, and 1:16 dilutions (each n=5), with recovery ranges of 87-91%, 87-107%, 74-101%, and 92-97% in serum; 90-105%, 84-101%, 90-101%, and 79-108% in EDTA plasma; and 84-95%, 92-105%, 82-105%, and 89-91% in heparin plasma, respectively.

An antibody specific for PRG2 is immobilised on the plate. Standards and samples are added and bound by the capture antibody, followed by washing and incubation with a Biotinylated Anti-PRG2 Antibody and HRP-Avidin conjugate. TMB substrate produces a blue colour that turns yellow upon addition of stop solution, with absorbance read at 450 nm on a colorimetric microplate reader. Components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
