Human Macrophage Inflammatory Protein 3 Beta ELISA Kit

Manufactured for Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human MIP-3 beta/CCL19 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay is reactive with human samples and targets Macrophage Inflammatory Protein 3 Beta.

The kit uses the sandwich enzyme immunoassay technique. An antibody specific for MIP-3 beta/CCL19 is pre-coated onto a 96-well microtiter plate. Standards and samples are added so that analyte present binds to the immobilised antibody. After washing, a Biotinylated Anti-MIP-3 beta/CCL19 Antibody binds the captured analyte, followed by an HRP-Avidin conjugate. TMB substrate produces a blue-coloured product that turns yellow upon addition of acidic stop solution; the density of yellow is proportional to the amount of MIP-3 beta/CCL19 captured in each well. Concentration is calculated by reading absorbance at 450 nm in a microplate reader and referring to the standard curve.

Key specifications include a product range of 15.6-1000 pg/ml and a sensitivity of <6.4 pg/ml. The pre-coated microplate comprises 12 x 8 well strips, and total assay time is 4h 30m. Detection is colourmetric. Kit components are stored at +4°C or -20°C as instructed; the kit should not be used past its expiration date. Reported sample recoveries averaged 91% for serum (range 83%-100%, n=5), 94% for EDTA plasma (range 91%-98%, n=5) and 94% for heparin plasma (range 90%-99%, n=5).
