A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is intended for the in vitro quantitative determination of human CCL4/MIP-1 beta in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique with an antibody specific for CCL4/MIP-1 beta pre-coated onto a 96-well microplate. Standards and test samples are added to the wells and the target is captured by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-CCL4/MIP-1 beta Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The absorbance is read at 450nm in a microplate reader and the concentration of CCL4/MIP-1 beta is calculated against a standard curve.

The product range is 15.6-1000 pg/ml, with a sensitivity of <5.5 pg/ml, and a total assay time of 4h 30m. The colourimetric readout is performed on a microplate reader. Recovery has been characterised across sample matrices, with an average recovery of 91% in serum (range 83-100%, n=5), 96% in EDTA plasma (range 90-103%, n=5), and 95% in heparin plasma (range 89-102%, n=5). Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. Reactivity is Human and the protein target is Macrophage Inflammatory Protein 1 Beta (CCL4/MIP-1 beta).
