Human Macrophage Colony Stimulating Factor ELISA Kit

From Antibodies.com comes a sandwich ELISA (sELISA) for the in vitro quantitative determination of human Macrophage Colony Stimulating Factor (M-CSF) in serum, plasma and cell culture supernatant samples. The assay is run on a pre-coated microplate (12 x 8 well strips) and read on a colorimetric microplate reader at 450 nm ± 2 nm.

The microplate is pre-coated with a monoclonal antibody specific to M-CSF. Samples or standards are added together with a biotin-conjugated polyclonal antibody preparation specific for M-CSF; any M-CSF present becomes immobilised by the coated antibody and is then sandwiched by the biotin conjugate. After washing, Avidin conjugated to Horseradish Peroxidase (HRP) is added, followed by a TMB substrate solution. The enzyme-substrate reaction is terminated with sulphuric acid solution and the colour change is measured spectrophotometrically. Two calibration diluents are supplied — Calibrator Diluent I for serum/plasma testing and Calibrator Diluent II for cell culture supernatant testing — and the standard is diluted 2-fold with the appropriate diluent to generate a standard curve of Optical Density versus M-CSF concentration in pg/mL.

Performance characteristics quoted by the manufacturer include an assay range of 120-4000 pg/ml, sensitivity of < 15 pg/ml and an assay time of 4h. The kit shows no cross-reactivity with other cytokines tested and detects both natural and recombinant human M-CSF. Reagents are shipped and stored at 4°C.
