From Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of human CD58, also known as Lymphocyte Function Associated Antigen 3, in serum, plasma, cell lysates and other biological fluids. The assay uses a pre-coated 96-well microtiter plate with colorimetric detection by absorbance measurement at 450 nm in a microplate reader.

In the sandwich enzyme immunoassay workflow, standards and samples are added to wells where CD58 is captured by the immobilized antibody. After washing, a biotinylated anti-CD58 detection antibody binds the captured analyte, followed by an HRP-avidin conjugate. TMB substrate produces a blue-coloured product that converts to yellow upon addition of acidic stop solution, with the intensity of the yellow colour directly proportional to the amount of CD58 captured in each well. Sensitivity is 5.8 pg/ml and the assay range is 12.5–800 pg/ml. The complete assay time is 4 hours 30 minutes, and recovery and dilution linearity have been validated for serum, EDTA plasma and heparin plasma samples.
