Human Lymphocyte Activation Gene 3 ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit from Antibodies.com enables the in vitro quantitative determination of human LAG-3 (Lymphocyte Activation Gene 3) in tissue homogenates, cell lysates, or other biological fluids. The quantitative assay uses a pre-coated 96-well microtiter plate and employs the sandwich enzyme immunoassay technique: standards and samples bind to an immobilised anti-LAG-3 capture antibody, followed by a biotinylated anti-LAG-3 detection antibody and an HRP-avidin conjugate. Signal is developed with TMB substrate, producing a blue-coloured product that becomes yellow after addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and LAG-3 concentration is calculated by referring to the standard curve.

Key specifications include a measuring range of 0.156-10 ng/ml and a sensitivity of 0.063 ng/ml. The assay is run on a colorimetric microplate reader with a total run time of 4 hours 30 minutes. Recovery has been validated in serum (n=5, range 80%-102%, average 91%), EDTA plasma (n=5, range 81%-100%, average 90%) and heparin plasma (n=5, range 80%-89%, average 84%). Kit components should be stored at +4°C or -20°C as instructed, and the kit is intended for use with human samples only.
