Manufactured by Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of human Low Density Lipoprotein Receptor (LDL Receptor) in tissue homogenates, cell lysates and other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) onto which an LDL Receptor-specific capture antibody has been immobilised. Standards and test samples are added, followed by a Biotinylated Anti-LDL Receptor Antibody and an HRP-Avidin conjugate. A TMB substrate is used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of the acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader, and LDL Receptor concentration is calculated from the standard curve.

Reported sensitivity is 0.118 ng/ml with an assay range of 0.312-20 ng/ml. Total assay time is 4h 30m. Recovery has been reported across serum (80-102%, average 91%), EDTA plasma (81-100%, average 90%) and heparin plasma (80-89%, average 84%) samples (n=5). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 has been reported as 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma (n=5).

Components supplied include a pre-coated 96-well microplate, lyophilised standard, Detection Solution A (120 μl), Detection Solution B (120 μl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers. Storage requirements vary by component between +4°C and -20°C.
