From Antibodies.com, this Human Liver X Receptor Alpha (LXR alpha) ELISA Kit is a sandwich enzyme-linked immunosorbent assay designed for the in vitro quantitative determination of human LXR alpha in serum, plasma, tissue homogenates or other biological fluids. An antibody specific for LXR alpha is pre-coated onto the microplate; standards and samples are added, followed by a biotinylated anti-LXR alpha antibody and an HRP-avidin conjugate. TMB substrate is then added and the enzymatic reaction is stopped with acidic stop solution, producing a yellow colour read at 450 nm on a colorimetric microplate reader and quantified against a standard curve.

The kit is for human reactivity and is suitable for serum, plasma and tissue homogenates. The stated assay range is 0.156–10 ng/ml and the sensitivity is <0.043 ng/ml, with a total assay time of 4 h 30 min. In spiked recovery testing (n=5), average recovery was 91% in serum (range 80–102%), 90% in EDTA plasma (range 81–100%) and 84% in heparin plasma (range 80–89%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions was 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma. Components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date.
