From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit is designed for the in vitro quantitative determination of human NR5A2/LRH1 (Liver Receptor Homolog 1) in tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique on a pre-coated microplate, with the target captured by an immobilised antibody and then detected using a Biotinylated Anti-NR5A2/LRH1 Antibody, HRP-Avidin conjugate, and TMB substrate. The enzymatic reaction produces a blue product that turns yellow after addition of acidic stop solution, with absorbance read at 450nm in a colorimetric microplate reader.

The kit reports a sensitivity of <0.112 ng/ml and an assay range of 0.312-20 ng/ml, with a total assay time of 4h 30m. Spike-and-recovery validation gave ranges of 80%-102% (average 91%) in serum (n=5), 81%-100% (average 90%) in EDTA plasma (n=5), and 80%-89% (average 84%) in heparin plasma (n=5). Linearity was assessed at 1:2, 1:4, 1:8, and 1:16 dilutions across serum, EDTA plasma, and heparin plasma samples (n=5).
