Description

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit from Antibodies.com with reactivity against human samples enables quantitative in vitro determination of human Lipoprotein A in serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated 96-well microplate supplied as 12 x 8 well strips, onto which an antibody specific for Lipoprotein A has been immobilised. Standards and test samples are added to the wells and the analyte is captured. After washing, a biotinylated anti-Lipoprotein A antibody is added, followed by an HRP-avidin conjugate and TMB substrate solution. The enzymatic reaction produces a blue product that turns yellow after addition of acidic stop solution; absorbance is read at 450 nm in a microplate reader and the Lipoprotein A concentration is calculated against the standard curve.

The kit is intended for use with a colorimetric microplate reader. The stated product range is 0.234–15 ng/ml with a sensitivity of <0.09 ng/ml. Total assay time is 4 hours 30 minutes. Reported overall recovery averages 95% in serum (range 89–101%, n=5), 96% in EDTA plasma (range 91–101%, n=5) and 96% in heparin plasma (range 91–101%, n=5). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 is reported as 83–96%, 90–104%, 74–103% and 90–93% for serum; 93–105%, 81–106%, 90–96% and 84–108% for EDTA plasma; and 79–98%, 92–101%, 86–105% and 87–96% for heparin plasma.

Kit components include the pre-coated microplate (12 x 8 well strips), lyophilised standard, detection solution A, detection solution B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers, with storage at +4°C or –20°C as instructed. The kit should not be used past its expiration date.
