The Antibodies.com Human Lipase, Gastric ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) intended for the in vitro quantitative determination of human LIPF in serum, plasma or other biological fluids. The assay uses the sandwich enzyme immunoassay technique, in which an antibody specific for LIPF is pre-coated onto a microtiter plate. Standards and test samples are added to the wells and captured LIPF is detected using a Biotinylated Anti-LIPF Antibody followed by an HRP-Avidin conjugate. A TMB substrate solution is then added; the HRP-catalysed reaction produces a blue product that turns yellow after addition of acidic stop solution, and absorbance is read at 450 nm in a microplate reader. The concentration of LIPF is calculated by reference to the standard curve.

According to the manufacturer's specifications, the kit has a sensitivity of <0.057 ng/ml and an assay range of 0.156–10 ng/ml. Total assay time is 4 hours 30 minutes, detection is colourimetric, and the platform is a pre-coated microplate configured as 12 x 8 well strips. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. The protocol is described as easy to follow.

Reported average recovery values (n=5) from the manufacturer are 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%), and 84% for heparin plasma (range 80–89%). Dilution linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16, with serum recoveries of 87–91%, 87–107%, 74–101% and 92–97% respectively; EDTA plasma recoveries of 90–105%, 84–101%, 90–101% and 79–108%; and heparin plasma recoveries of 84–95%, 92–105%, 82–105% and 89–91%.
