Description
From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human LAT (Linker For Activation Of T Cell) in tissue homogenates, cell lysates, and other biological fluids. The assay uses a pre-coated microplate format, with absorbance read at 450nm on a colorimetric microplate reader.
An antibody specific for LAT is immobilised on the plate to capture the target from standards and samples, followed by incubation with a biotinylated anti-LAT detection antibody. An HRP-Avidin conjugate is then added and TMB substrate is used to visualise the enzymatic reaction, producing a blue product that changes to yellow upon addition of the acidic stop solution. The intensity of the yellow colour is proportional to the amount of LAT captured, and the concentration is calculated from a standard curve. Total assay time is 4h 30m.
Sensitivity is <0.102 ng/ml with an assay range of 0.312-20 ng/ml. Reported recovery (n=5) ranges were 80%-102% for serum (average 91%), 81%-100% for EDTA plasma (average 90%), and 80%-89% for heparin plasma (average 84%). Linearity for serum (n=5) across dilutions was 87-91% at 1:2, 87-107% at 1:4, 74-101% at 1:8, and 92-97% at 1:16. Kit components should be stored at +4°C or -20°C as instructed.
