From Antibodies.com, this Human LASP1 ELISA Kit is a sandwich enzyme-linked immunosorbent assay (sELISA) designed for the in vitro quantitative determination of human LASP1 in tissue homogenates, cell lysates, and other biological fluids. The kit is reactive with human samples and is intended for research use only.

The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added to wells where LASP1 is captured by the immobilised antibody. Following incubation and washing, a biotinylated anti-LASP1 antibody is added, followed by an HRP-avidin conjugate. Signal is visualised using TMB substrate, producing a blue colour that turns yellow upon addition of the acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader, and LASP1 concentration is calculated by reference to a standard curve. The kit reports a sensitivity of 0.065 ng/ml, an assay range of 0.156–10 ng/ml, and a total assay time of 4 hours 30 minutes.

Validated sample types include serum, EDTA plasma, heparin plasma, tissue homogenates, cell lysates, and cell culture supernatant. Reported recovery (n=5) is 80%–102% (average 91%) for serum, 81%–100% (average 90%) for EDTA plasma, and 80%–89% (average 84%) for heparin plasma. Linearity data (n=5) at dilutions of 1:2, 1:4, 1:8, and 1:16 are also provided for each matrix. Kit components should be stored at +4°C or –20°C as instructed, and the product should not be used past its expiration date.
