Human Leptin ELISA Kit

This Antibodies.com sandwich ELISA (sELISA) is designed for the in vitro quantitative determination of human Leptin in serum, plasma, saliva, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids. The microplate is pre-coated with an antibody specific for Leptin, which captures the analyte from standards and test samples. After incubation and washing, a Biotinylated Anti-Leptin Antibody is added and binds the captured Leptin; HRP-Avidin conjugate is then introduced. The HRP enzymatic reaction is visualised with TMB substrate, producing a blue-coloured product that changes to yellow after the addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and Leptin concentration is calculated against a standard curve.

The assay is run on a pre-coated microplate with colourimetric detection and a total assay time of 4 h 30 m. Reported sensitivity is <0.059 ng/ml and the product range is 0.156-10 ng/ml. Recovery has been assessed in five replicates per matrix: serum 80-102% (average 91%), EDTA plasma 81-100% (average 90%) and heparin plasma 80-89% (average 84%). Kit components are stored at +4°C or -20°C as instructed and must not be used past the expiration date.
