Human Legumain ELISA Kit

From Antibodies.com, this Human Legumain ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of human Legumain in serum, plasma, tissue homogenates, cell lysates or other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. An antibody specific for Legumain is immobilised on the plate to capture the analyte, followed by a Biotinylated Anti-Legumain Antibody and an HRP-Avidin conjugate. TMB substrate is then added to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow after addition of the acidic stop solution. The absorbance is read at 450 nm on a colorimetric microplate reader and the Legumain concentration is calculated by reference to a standard curve.

The kit has a stated sensitivity of 0.118 ng/ml and a quantitative product range of 0.312–20 ng/ml, with a total assay time of approximately 4 hours 30 minutes. Reported spike-and-recovery averages (n=5) are 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%) and 84% for heparin plasma (range 80–89%). Dilutional linearity for n=5 samples at 1:2, 1:4, 1:8 and 1:16 falls within 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma. Kit components are stored at +4°C or −20°C as instructed in the protocol, and the product is for research use only.
