Human Left/Right Determination Factor 1 ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of human Left/Right Determination Factor 1 (Lefty) in tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay employs the sandwich enzyme immunoassay technique on a pre-coated microplate and is read on a colorimetric microplate reader at 450 nm.

An antibody specific for Lefty is pre-coated onto the microplate. Standards and samples are added to the wells and Lefty present in each sample is captured by the immobilized antibody. After washing, a Biotinylated Anti-Lefty Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualize the reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Lefty captured, and the concentration of Lefty is calculated by referring to the standard curve.

The product range of the assay is 0.781–50 ng/ml, with a reported sensitivity of <0.24 ng/ml. Total assay time is approximately 4 hours 30 minutes. Kit components are stored at +4°C or -20°C as instructed. Recovery in serum (n=5) was 80–102% (average 91%); in EDTA plasma (n=5), 81–101% (average 91%); and in heparin plasma (n=5), 80–89% (average 84%). The kit should not be used past its expiration date.
