The Antibodies.com sandwich ELISA kit is designed for the in vitro quantitative determination of human Latrophilin 3 (LPHN3) in tissue homogenates, cell lysates and other biological fluids. The assay uses a pre-coated microplate immobilised with an antibody specific for LPHN3. Standards and test samples are added to the wells, followed by a biotinylated anti-LPHN3 antibody and an HRP-avidin conjugate. A TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of an acidic stop solution. The density of yellow is proportional to the amount of LPHN3 captured in each well and is read at 450 nm in a microplate reader, with concentration calculated against the standard curve.

Reported performance figures include a sensitivity of <0.114 ng/ml, an assay range of 0.312–20 ng/ml and a total assay time of 4h 30m. Spike-and-recovery was assessed in serum (n=5) with a range of 88–101% and an average of 94%; in EDTA plasma (n=5) with a range of 91–103% and an average of 97%; and in heparin plasma (n=5) with a range of 92–98% and an average of 95%. Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 across serum, EDTA plasma and heparin plasma samples. Kit components are stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
