Human Laminin Beta 2 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com for the in vitro quantitative determination of human Laminin beta 2 in serum, plasma, tissue homogenates or other biological fluids. The assay runs on a pre-coated 96-well microplate (12 x 8 well strips) bearing an immobilised antibody specific for Laminin beta 2. Standards and samples are added to the wells, the plate is washed, and a biotinylated anti-Laminin beta 2 antibody is added followed by HRP-avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to Laminin beta 2 captured in each well, and concentration is calculated by reading O.D. absorbance at 450 nm against the standard curve.

The assay range is 0.781-50 ng/ml with a sensitivity of 0.33 ng/ml, read on a colorimetric microplate reader, and total assay time is 4h 30m. Reported average recoveries are 91% in serum (n=5, 83%-99%), 89% in EDTA plasma (n=5, 80%-99%) and 90% in heparin plasma (n=5, 82%-99%), with dilution linearity assessed at 1:2, 1:4, 1:8 and 1:16. Supplied components include a lyophilised standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers (5 adhesive strips), to be stored at +4°C or -20°C as instructed.
