Human Lamin B1 ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of human Lamin B1 in tissue homogenates, cell lysates, or other biological fluids. A capture antibody specific for Lamin B1 is pre-coated onto the microplate wells. Standards and test samples are added to the wells; after incubation and washing, a Biotinylated Anti-Lamin B1 Antibody binds the captured analyte. Following addition of an HRP-Avidin conjugate, TMB substrate is used to visualise the enzymatic reaction, producing a blue colour that changes to yellow upon addition of the stop solution. Absorbance is read at 450 nm in a microplate reader, and Lamin B1 concentration is calculated from the standard curve.

The assay has a quantitative range of 0.312-20 ng/ml and a sensitivity of <0.118 ng/ml. The total run time is 4h 30m, and the assay is read on a colorimetric microplate reader. Kit components are stored at +4°C or -20°C as directed, and the kit must not be used past its expiration date.

Spike-and-recovery testing in serum (n=5) gave an average of 91% across a range of 80%-102%. EDTA plasma (n=5) returned an average of 90% (range 81%-100%), and heparin plasma (n=5) averaged 84% (range 80%-89%). Linearity of dilution in serum (n=5) at 1:2, 1:4, 1:8 and 1:16 returned 87-91%, 87-107%, 74-101% and 92-97%; EDTA plasma (n=5) gave 90-105%, 84-101%, 90-101% and 79-108%; heparin plasma (n=5) gave 84-95%, 92-105%, 82-105% and 89-91%.
